HRP Chromogen for IHC - DAB
DAB (3,3'-Diaminobenzidine) is the most widely used chromogen for use with HRP. DAB substrate forms a brown colored deposit when it reacts with peroxidase in the presence of hemoglobin.
The use of DAB is simple, simply incubate the slides for 5 minutes in a humid chamber at room temperature. The appearance of the staining can be followed by observation under the microscope.
When staining is complete, it is necessary to rinse the slides with water for 1-2 minutes to remove excess DAB. The precipitate from DAB is insoluble in alcohol and other organic solvents. Samples can be dehydrated and slides mounted permanently (O-Mount).
