  • Western blot analysis of extracts of A-549 cells, using ICAM-1/CD54 antibody at 1:1000 dilution. A-549 cells were treated by TNF-Alpha (20 ng/mL) at 37 °C for 30 minutes. Secondary antibody: HRP Goat Anti-rabbit IgG (H+L) at 1:10000 dilution. Lysates/proteins: 25 Mu g per lane. Blocking buffer: 3% BSA. Detection: ECL Enhanced Kit. Exposure time: 3min.
  • Immunohistochemistry analysis of paraffin-embedded Human lung cancer using ICAM-1/CD54 antibody at dilution of 1:100 (40x lens). Perform microwave antigen retrieval with 10 mM PBS buffer pH 7. 2 before commencing with immunohistochemistry staining protocol.
  • Flow cytometry:1X10^6 293F cells (negative control, left) and Raji cells (right) were surface-stained with ICAM-1/CD54 rabbit monoclonal antibody or rabbit IgG isotype control (10 Mu g/mL, blue line) , followed by Alexa Fluor 647 conjugated goat anti-rabbit polyclonal antibody (1:600 dilution) staining. Non-fluorescently stained cells were used as blank control (red line).

Function ICAM proteins are ligands for the leukocyte adhesion protein LFA-1 (integrin alpha-L/beta-2). During leukocyte trans-endothelial migration, ICAM1 engagement promotes the assembly of endothelial apical cups through ARHGEF26/SGEF and RHOG activation. (Microbial infection) Acts as a receptor for major receptor group rhinovirus A-B capsid proteins. (Microbial infection) Acts as a receptor for Coxsackievirus A21 capsid proteins. (Microbial infection) Upon Kaposi's sarcoma-associated herpesvirus/HHV-8 infection, is degraded by viral E3 ubiquitin ligase MIR2, presumably to prevent lysis of infected cells by cytotoxic T-lymphocytes and NK cell.
